I guess they had some sort of memory scanning tool open and they knew what address of code would be run after the glitch happened, then they figured out how to manipulate the bits around that address using lots of (tool assisted) trial and error.

AGDQ 2015 Ninja Gaiden Relay Race, involving multiple world record holders in a Ninja Gaiden 1, 2 and 3 continuous race. It's incredible and way more intense than I imagined it would be, I'm halfway through watching it and it's such an incredible competition, with two excellent commentators who point out what's happening and explain things really well, which is key for an interesting live speedrun.
