There's actually nothing more in-jokey to it than that. The Mad Duck is an iconic Earthbound enemy because of how pitifully weak and goofy-looking it is. Someone took the picture and made it shake its head like that one famous bird. Then someone did the siren-eyes thing for laughs. That's all there is to it.
