In RE4, you don't need to do anything special to learn the suplex. For knee stuns, Leon uses the suplex on zealot/militia Ganados and a back kick on villagers. You can test this by going back inside the church where you first rescue Ashley after jumping out the window. There are 2 zealots hidden there, before you get to the castle you can use the suplex on them. And in the mines, the knee stun on villager Ganados gives the KICK prompt, not the suplex.
