The hell, did EO V really just come out? I bought it on the eShop Thursday on a whim after not playing an EO since like one floor of the first game and twenty or so hours of P:Q. Currently running:

Fencer/Harbinger
Warlock(I think?)/Rover/Herbalist

A couple of dumb questions:
-How often should I expect to rest/retire/whatever characters and swap them out? Or is it better to just use a party the whole way through?
-Do the increasing Inn costs ever become a problem? I'm worried about boning myself by playing conservatively instead of pushing the limits every time I go to the dungeon.
-How many floors are there (or is that a spoiler? Never gotten far enough in one of these to know)?

1. Really depends on how you feel or if you need to try a new strat. Resting really isn't a big deal. There's also an XP share accessory in a hidden spot on the 3rd floor and some items that level a character up to 20 you get after the second boss. I really like to swap out party members and play with different comps but most people settle on a particular core group eventually. Some classes (harbinger and shaman mainly) are pretty thirsty for skill points so some people swap them into their parties later on, and some parties are only really going to work when the specialisations get unlocked. There's some bosses in the postgame you might want to change your setup for. Just do what you feel like and keep that memory conch on somebody.

2. Inn costs are never really a big deal, if it's an issue go clown on an early floor and sell bits. The costs of everything else can get annoying though.

3. There's 6 stratums (layers) with 5 floors each, one layer is postgame stuff though.

Single sword masurao should be pretty good, a lot of their skills are kinda eh but Helm Splitter is pretty cool, and they’d be real survivable since they get heavy armour and that counterattack skill. They have some interesting synergies since they actually have a low accuracy attack so they’ll buddy up with pugilists and Rovers for leg binds, or somebody with Petrify.

Do I need to worry about Race/Class combinations other than the defaults? I'm pretty happy with the way my team's looking in this First Stratum with their default Races, but if late game isn't feasible without "optimal" stuff I'd rather know to change it now then later.

The only one I’d recommend a normal play through is if you want a smoke-focused botanist, reclass a celestrian. Everything else should be fine though unless you’re trying to one-shot the super boss.

Really depends on the party setup but it’s only worth worrying about with poison.

Depends if you’re going for a chain killer one or just elemental chains.

Alternatively, pull a Warrior Might and cheese it with 5 Dancers. Only it's actually reliable and not the worst cheese method to use, unlike in EO3.

I'm a little surprised that dancers work so well. I never really got the hang of them aside from as a fortress subclass.

I'm starting up my old EO5 file and I'm trying to rejig my party where I left off at the early 3rd stratum. I had the standard party of

Gun Dragoon
Binds Cestus
6 Element Wizard
super healer botanist
and a hound rover

but I'm finding the whole thing a little lacking in damage. The weakest link is the dog dude, he doesn't output much damage or heals, and the cestus handles binds well enough. I don't know what to replace him with though. A masurao could work but 4 swords is too fragile without adding more lockdown stuff, and IDK if a single sword one is worth it. I tried a reaper but they're too fiddly for random encounters and I don't want to grind. The super healer might be overkill but I'm not sure about a replacement either. Is a shaman going to be good enough? Is there anything I'm missing?

Have you considered a bird dude? Hound is mostly a healing spec, so it's obviously redundant if you have a healer botanist. Bird does massively more damage.

I’m rolling with the bird now and it’s good

you explicitly end up in Tokyo in EO1, don't get too caught up with geography.

I could use a quick opinion before jumping into EO; I've never played any of these, and have been informed that IV is probably the bestest one. Is it worth playing III first, so I can enjoy the best of the DS ones before I see all the QoL improvements the 3DS games had? Or should I just jump directly into IV?

definitely start with 4, 3 is kind of busted in a lot of ways and you need a level of system mastery to really get the most out of it

VFD is about the weirdest a game can be while still being insanely cliche. I appreciate the bonkers classes and extended party stuff though.
